Metachromasia with basic thiazine dyes such as toluidine and methylene blue results ina color shift from blue to deep purple . This metachromatic shift is due to the binding of dye molecules ina tight array on polyanionic molecules such as glycosamminoglycans(GAG) . In the blood , the basophil is the only cell that contains large metachromatic granules. This reaction occurs because the granules are rich in the GAG heparin.
