Dopamine itself was tried, but it did not cross the BBB. Dopamine’s immediate precursor, L-dihydroxyphenylalanine (L-DOPA), was then used instead. L-DOPA is actively transported into the brain by the neutral amino acid transporter and, once peripheral metabolism was blocked, it was used to great effect in the treatment of Parkinson’s disease.
